Application Notes | The mAb clearly inhibits signalling by Notch, and also Eph receptors, however the antibody does not interact directly with residues identified as contributing to substrate binding. The mAb recognises a specific conformation of the ADAM10 substrate binding domain dependent on CxxC bonding. The mAb binding is blocked by mutation of the CxxC motif, and is altered by modulating the redox environment. mAb-immunoprecipitates of ADAM10 showed highly significant enrichment of protease activity. Also, oxidative conditions, known to enhance ADAM activity, favoured the mAb binding. |
