Description | This product is a broadly neutralizing antibody against NiV F. The mAb is derived from a mouse antibody. The antibody binds to a quaternary epitope on domain III of the NiV F globular head. 5B3 effectively inhibits NiV and HeV infection of target cells. |
Clonality | Monoclonal |
Host Species | Mouse |
Target Species | Nipah virus (NiV), Hendra virus (HeV) |
Epitope | domain III of the F glycoprotein |
Neutralization Mechanism | 5B3 locks F in the prefusion conformation and prevents membrane fusion via molecular stapling, providing a molecular rationale for its potency. |
Isotype | Mouse IgG1 |
Expression Species | HEK293F or CHO |
Conjugation | None |
Purity | >95%, determined by SDS-PAGE and SEC-HPLC |
Endotoxin | <1 EU/mg |
Purification | Protein G purified |
Sterility | 0.2 μM filtered |
Formulation | PBS, pH 7.4 |
Preservation | No preservatives |
Stabilizer | No stabilizers |
Storage | Store at 4°C within a week. For longer storage, aliquot and store at -20°C. |
Application | ELISA; Neut; IP |
Application Notes | The half-maximal inhibitory concentrations (IC₅₀) were determined to be 1.2 µg/mL. NiV mutants resistant to neutralization were created by incubating 1 x 10⁵ 50% tissue culture infective dose of each virus with a sub-neutralizing amount of 40 μg of 5B3 IgG in 100 μl of medium for 1 hour at 37°C. The mixture was then inoculated onto 1 x 10⁶ VeroE6 cells in the presence of IgG at the same concentration. |
