Description | The antibody XG014 is a human mAb specific for SARS-CoV-2 Spike S1 protein. The antibody is a receptorbinding domain (RBD)-binding antibody, which potently neutralizes β-coronavirus lineage B (β-CoV-B), including SARS-CoV-2, its circulating variants, SARS-CoV and bat SARSr-CoV WIV1. It recognizes a conserved epitope outside the ACE2 binding site and completely locks RBD in the non-functional "down" conformation. |

Application Notes | ELISA plates were coated with 10 µg/mL of S-ECD or RBD of wild-type SARS-CoV-2, or its variants, including B.1.1.7, B.1.351 and P.1 in PBS overnight at 4°C. Plates were then blocked with 2% BSA in PBS. Antibodies were three-fold serially diluted with a maximum concentration of 10 µg/mL for eight dilutions and incubated for one hour at room temperature. Detection was performed after incubating with HRP-conjugated goat anti-human IgG for one hour at room temperature. The area under the curve (AUC) was calculated by using PRISM software. |
