Description | The antibody F61 is a human mAb specific for SARS-CoV-2 Spike S1 protein. It exhibits high-affinity binding to Delta-RBD, Omicron-RBD-BA.1 and Omicron RBD-BA.2. The KD values to Delta-RBD, Omicron-RBDBA.1 and Omicron-RBD-BA.2 by the antibody were ~3.89 pM, ~5.03 nM and ~93.3 pM, respectively. It was highly potent in neutralizing SARS-CoV-2 WT pseudovirus with half-maximal inhibitory concentration (IC50) of 6 ng/mL. It exhibited high potency and broad neutralization against Alpha, Beta, Delta and Omicron, even its binding to Omicron-RBD-BA.1 was significantly reduced (~1000-fold) compared to WT-RBD. |

Application Notes | The antibody was highly potent in neutralizing SARS-CoV-2 WT pseudovirus with half-maximal inhibitory concentration (IC50) of 6 ng/mL. ELISA plates were coated with SARS-CoV-2 protein including WT-S1, WT-NTD, WT-RBD, Delta-S1, Delta_x0002_RBD and Omicron-RBD at 4°C overnight. Following washing with PBST, serial dilutions of testing antibodies starting at 5 μg/mL were added to each well and incubated at 37°C for 30 min. After washing with PBST, HRP-conjugated anti-human IgG Fc specific antibody was added at the dilution of 1:2000 and incubated at 37°C for 30 min. The absorbance was detected at 450 nm. |
