Angiopoietin 1 (ANGPT1), also known as Ang1 or KIAA0003, is a crucial member of the angiopoietin family. ANGPT1 has a potential fibrinogen-like domain and a special coiled-coil region. ANGPT1 is primarily found in the lung and myocardium of the atrium and ventricles. In adults, ANGPT1 is also found in the heart and liver. As a secreted glycoprotein, ANGPT1 activates target receptors by inducing tyrosine phosphorylation. ANGPT1 is involved in regulating interactions between the surrounding matrix and the endothelium. ANGPT1 is also a key regulator of endothelial permeability.

The receptor system of ANGPT1 consists of two receptor tyrosine kinases, Tie1 and Tie2. It is reported that ANGPT1 signals through Tie1 and Tie2 and is involved in the regulation of a variety of life activities.

There is some evidence revealing that ANGPT1 binds to several integrins in addition to Tie1 and Tie2, including α2β1, α5β1, αvβ3, and αvβ5.
ANGPT1 is an indispensable regulator in vascular development, angiogenesis, and vascular remodeling. Furthermore, ANGPT1 protects blood vessels by inhibiting plasma leakage, suppressing vascular inflammation, and preventing endothelial cell death. Substantial evidence indicates that ANGPT1 plays a critical role in facilitating vascular maturation and stability. Recent studies suggest that ANGPT1 has a potential therapeutic role in specific conditions, including the treatment of endotoxemia, edema, and graft atherosclerosis.
The ANGPT1-Tie signaling pathway is closely related to the health of the cardiovascular system. Dysregulation of the ANGPT1-Tie signaling axis may lead to vascular damage and the formation of atherosclerotic plaques. Additionally, abnormalities in this pathway may lead to peripheral arterial disease. Notably, ANGPT1-Tie is a key regulator of neovascularization and vascular leakage in some eye diseases.
