Angiopoietin 2 (ANGPT2), also known as LMPHM10 or Ang2, is a pivotal member of the angiopoietin family. ANGPT2 is mainly produced by endothelial cells and contributes to angiogenesis. Generally, ANGPT2 is present at low levels in tissues. After birth, ANGPT2 is expressed only in highly vasculogenic tissues, such as the ovary and uterus. However, ANGPT2 was shown to be highly upregulated in tumor vasculature. In addition, ANGPT2 has been found to be up-regulated in several inflammatory diseases and is directly involved in the regulation of inflammation-related signaling pathways.

Angiopoietin 1 (ANGPT1) and ANGPT2 are both vital ligands for the endothelial TEK receptors. ANGPT1 is highly homologous to ANGPT2, and both bind to the same site on Tie2. Therefore, ANGPT2 is regarded as an antagonist of ANGPT1. ANGPT2 has been reported to adversely affect the vascular remodeling function of ANGPT1 and may promote apoptosis of endothelial cells. It is worth noting that ANGPT2 is also an indispensable regulator of postnatal vascular remodeling, and it cooperates with ANGPT1 during lymphatic development. ANGPT2 is also closely associated with angiogenesis during embryogenesis and tumorigenesis.

Increasing evidence shows that ANGPT2 is highly expressed in various tumor cells and involved in the regulation of tumor angiogenesis and inflammation. Abnormal levels of ANGPT2 are closely linked to the occurrence, development, and prognosis of cancer. It has been reported that abnormal expression of ANGPT2 may lead to the occurrence of a variety of cardiovascular diseases, including congestive heart failure and coronary artery disease. In addition, the overexpression of ANGPT2 is significantly related to chronic kidney disease.
ANGPT2 has been validated as a potential biomarker for immunotherapy. Targeting ANGPT2 provides an attractive combinatorial approach to promote the efficacy of immunotherapy.
