Beta-secretase 1 (BACE1), also known as β-secretase, is a type I transmembrane protease. BACE1 cleaves the membrane-bound APP at the N-terminus to extracellularly release the soluble APP. BACE1 is associated with Alzheimer's Disease (AD) and Myositis.

Defective Signaling Pathways of BACE1

To study the molecular basis of BACE1-associated pathological generation and progression of AD, we focused on the defective signaling pathways of BACE1.

  • BACE1 promotes the production of insoluble Aβ isoforms.
  • NF-κB activation increases BACE1 expression, Aβ production, and downstream signaling pathways.
  • GSK3β triggers the activation of NF-κ, sequential BACE1 expression and Aβ production.

Therapeutic Strategies for BACE1

BACE1 activity is a key factor for the pathogenesis of AD, which highlights the potential of BACE1 as a promising target for the diagnosis and treatment research of AD. The BACE1-related strategies are listed below:

  • The concentration level and the enzymatic activity of BACE1 could be monitored as biomarkers for the treatments of AD by targeting BACE1.
  • Small-molecule inhibitors of BACE1 could be developed to lower the insoluble Aβ levels.
  • BACE1-based bispecific antibodies that could penetrate the blood-brain barrier may alleviate the dysfunction and progression of AD pathology. As shown in Fig.2, the transferrin-BACE1 bispecific antibody guarantees higher antibody working concentrations in the brain and enhanced therapeutic responses.
