CD200R1 (Cell surface transmembrane glycoprotein CD200 receptor 1, also known as OX2R), is an Ig superfamily transmembrane glycoprotein containing two immunoglobulin-like domains. CD200R1 is primarily distributed on the surface of CD4+ T cells, CD8+ T cells, and myeloid cells. It interacts with its foremost ligand, CD200 transmembrane glycoprotein, to repress the functions of myeloid cells, the latter is expressed on a broad spectrum of cells including epithelial cells, neurons, endothelial cells, lymphoid cells, and fibroblasts. CD200R1 is closely involved in the negative regulation of multiple immune responses, which facilitates the prolongation of survival of transplanted allografts, while it also leads to weakened resistance to decreased resistance to tumor growth in some cases.

CD200r identifies and interacts with the ligand CD200, and triggers downstream signaling pathways like tyrosine kinase (Dok) 1 and Dok2. While Dok1 recruited less RasGAP than Dok2, Dok2 recruited the adaptor molecule Nck following ligand engagement of CD200R in return, further leading to recruitment of RasGAP and SHIP, which diminishes the activation of p38 MAPK, MAPKs ERK, and JNK via the inactivating Ras. The mechanism thus affects MAP kinases, resulting in subsequent inhibition of Ras/MAPK activation, and suppressing the signaling of cytokine and pattern recognition receptors.

Activation of CD200R1 also participates in regulating of multiple pro-inflammatory molecules, such as tumor necrosis factor (TNF), inducible nitric oxide synthase, and interferons. For example, CD200:CD200R1-mediated signal inhibition is associated with downstream augmentation of the production of TGFβ and IL-10 molecules, leading to immunosuppression.
Antibodies targeting therapeutic targets has exhibited strong antitumor activities and has facilitated in the increase of survival in numerous cancers. As an immune inhibitory receptor, CD200R1 plays essential roles involved with the production of proinflammatory cytokines by T cells and myeloid cells, as well as the immune responses in autoimmune diseases or against infectious agents, making it a potential target of antibody development for research, clinical or commercial purposes.
In a recent study, CD200R1-blocking antibodies are utilized to inhibit the CD200:CD200R1 signaling pathway, revealing that suppressed CD200:CD200R1 signaling in psoriasis enhances neutrophil recruitment to the skin. Another study introduces a humanized anti-CD200R1 antibody with high binding affinity, which successfully blocks CD200 binding, down-regulating the recruitment of DOK2, leading to induction of T-cell cytokine production and enhanced cell killing, and significantly reducing tumor growth in a melanoma model in mice.
