CD27 Specific Neutra™ Antibody Products
Product listCD27 molecule (CD27), also known as TNFRSF7 or Tp55, is a single-pass type I membrane protein overexpressed in lymphoid tissues such as the spleen and peripheral blood mononuclear cells. As an essential receptor for T-cell immunity, membrane-bound CD27 binds to the ligand CD70 and plays a key role in regulating cellular anti-tumor immunity. Soluble CD27 also has effects on T cell activation and its expression level is considered as a biomarker of immune responses.

Functions of the CD27-CD70 Signaling Pathway
TLR- and CD40-induced expression of CD70 regulates the CD27-CD70 interaction and stimulates the NF-κB, JNK, PI3K, and MEK signaling pathways. The signal transduction may have the functions listed below (figure 1).
- Regulates activation of B cells and immunoglobulin synthesis.
- Promotes activation of T cells and their differentiation into T helper 1 cells.
- Transduction of costimulatory signals with TCR engagement.

The Role of the CD27-CD70 Pathway in Immune Evasion
The CD27-CD70 axis has bidirectional functional immune effects and may retrograde anti-tumor signals. As shown in Figure 2, the axis facilitates immune evasion and suppression of solid tumor cells and subsequently promotes tumor migration and invasion in the following ways:
- Increases survival, proliferation, and infiltration of regulatory T cells.
- Promotes T cell exhaustion and apoptosis.
- Enhances NK cell exhaustion.
- Recruits or activates tumor-associated macrophages (TAMs).

CD27 in Anti-tumor Immunotherapies
CD27 has been implicated in many novel anti-cancer studies. With a relatively comprehensive understanding of the CD70-CD27 signaling axis, CD27-related biological products could be used in many treatment strategies:
- Development of agonistic monoclonal antibodies for potential therapy of CD27 overexpressing lymphoma and leukemia.
- Administration along with anti-PD-1/PDL1 therapy in solid tumors.
- Implementation of CD27 in chimeric antigen receptor T cells (CAR-T) for optimal cell immunotherapy.
