The CD3E molecule, also known as CD3 epsilon, is a subunit of the CD3 complex, a critical component of the T cell receptor (TCR) complex. CD3e is primarily expressed on the surface of T cells, where it associates with other CD3 subunits (CD3γ, CD3δ, and CD3ζ) and the TCR α and β chains to form the TCR-CD3 complex. This complex functions as a signal transduction module that detects antigenic peptides presented by major histocompatibility complex (MHC) molecules on antigen-presenting cells. CD3E is essential for T cell development, activation, and function, making it a critical target for immunotherapy and a key molecule in the study of T cell biology and immune responses.

CD3E plays a central role in T cell activation by transmitting signals from the TCR to intracellular signaling pathways upon antigen recognition. It contains immunoreceptor tyrosine-based activation motifs (ITAMs) in its cytoplasmic tail, which are phosphorylated upon TCR engagement, leading to the recruitment and activation of downstream signaling molecules such as ZAP-70 and PI3K. This initiates a cascade of events culminating in T cell activation, proliferation, and effector functions, including cytokine production and cytotoxicity.
CD3E is involved in the T cell receptor complex, crucial for adaptive immune responses, which may influence its role in moderating asthma symptoms and response to treatment. Some researchers found that it showed a notable difference in expression between responders and non-responders. Then, CD3E was identified as a significant biomarker for predicting response to omalizumab through logistic regression analysis. The findings suggest that measuring CD3E levels could help identify which asthma patients are likely to benefit from omalizumab treatment, potentially leading to more personalized and effective asthma management.

Using human CD3E gene to replace mouse Cd3e gene to establish humanized CD3E mice can construct an effective tool for CD3E targeted drug efficacy evaluation and toxicological evaluation. These models contribute to advancing our understanding of cancer immunotherapy and facilitate the development of novel treatment strategies.
