Overview of CEACAM1

CEACAM1, also known as carcinoembryonic antigen-related cell adhesion molecule 1, is a transmembrane glycoprotein that plays important roles in various cellular processes, including cell adhesion, migration, and signaling.

  • Expression

CEACAM1 belongs to the CEACAM family, which is made up of glycoproteins that have similar structures. It is expressed on the surface of several cell types, such as epithelial cells, leukocytes, and endothelial cells.

  • Signaling pathway

CEACAM1 is involved in various signaling pathways, such as the PI3K-Akt, MAPK, and NF-κB pathways. These pathways regulate a variety of biological activities, including cell survival, proliferation, and inflammation.

  • Related diseases

Research has shown that CEACAM1 has been linked to several kinds of diseases, including cancer, inflammatory bowel disease, and bacterial infections. In cancer, CEACAM1 expression frequently changes, which can lead to tumor growth and metastasis. Inflammatory bowel disease patients have been shown to have decreased CEACAM1 expression in their intestinal tissue. In bacterial infections, CEACAM1 can act as a receptor for certain pathogens, facilitating their entry into host cells.

CEACAM1 Antibody for Cancer Therapy

CEACAM1 is often overexpressed in various cancers, including colorectal, pancreatic, and lung cancer. Targeting CEACAM1 with monoclonal antibodies or antibody-drug conjugates (ADCs) has shown promise in preclinical and early clinical studies. resulting in their death while preserving healthy cells. CEACAM1-specific neutralizing antibodies are designed to specifically target and neutralize CEACAM1 which has been shown to exhibit high specificity and affinity for CEACAM1. Preclinical studies have provided encouraging findings. In one study, therapy with the antibody results in considerable tumor regression in a pancreatic cancer animal model. The antibody also enhances the activity of standard chemotherapy agents, leading to further tumor suppression.

Related Therapies Targeting CEACAM1

CEACAM1 has been associated with the development of a variety of autoimmune disorders. Some studies have suggested that modulating the expression or function of CEACAM1 could be a potential therapeutic strategy for these diseases. Therapeutic strategies targeting CEACAM1 can be used in inflammatory bowel disease, aiming to restore its expression and function in the intestinal epithelium. EACAM1 also serves as a receptor for various pathogens, so developing CEACAM1-targeted therapies could potentially prevent or treat infections caused by these pathogens, such as blocking CEACAM1-pathogen interactions or disrupting bacterial adherence.
