Fibroblast growth factor receptor 1 (FGFR1), also known as CEK or FLG, is a membrane protein that is overexpressed in breast carcinomas and normal tissue like plasma and heart. FGFR1 is a receptor for fibroblast growth factors that transduce the signals regulating cell proliferation, differentiation, and migration.

Functions of FGFR1 Pathway

Overexpression of FGFR1 could promote the proliferation of lymphoma cells. Via the MYC-KDM2B-EZH2 cascade, FGFR1 regulates the expression of CDKN1C and E2F target genes. Subsequently, FGFR1 maintains the expression level of MYC and provides a proliferative advantage to lymphoma cells.

Treatment Strategies Targeting FGFR

The development of FGFR inhibitors is an important for FGFR related cancer studies. Depending on the mechanism of action, FGFR inhibitors can be classified into four types:

FGF ligand traps that block the activity of FGF-FGFRs interaction.
Antibody-drug conjugates (ADCs) that comprise a cytotoxic payload linking to an FGFR antibody.
Antagonistic anti-FGFR antibodies that block the activation of FGF-FGFRs signaling.
Small molecule tyrosine kinase inhibitors that non-selectively or selectively targets the kinase domain of FGFRs.
