Introduction to IL10

Interleukin 10 (IL10), a pleiotropic cytokine primarily secreted by regulatory T cells, macrophages, and B cells, plays a critical role in suppressing pro-inflammatory responses and maintaining immune homeostasis. It modulates both innate and adaptive immunity by inhibiting antigen presentation, Th1 cytokine production, and macrophage activation. Dysregulated IL10 signaling is implicated in pathologies ranging from autoimmune diseases to cancer immune evasion.

IL10 is a homodimeric protein composed of two 160-amino-acid subunits, each containing six α-helices arranged in a conserved helical bundle fold. Structural studies reveal that the C-terminal domains of IL10 mediate receptor binding, while the N-terminal regions stabilize dimerization—a key feature for biological activity.
