Product listInterleukin 17A (IL17A), also known as IL-17 or CTLA8, is an effective cytokine involved in antimicrobial host defense and tissue integrity protection. IL17A interacts with the IL17RA-IL17RC receptor complex and triggers a series of downstream pathways. IL17A is associated with inflammatory, autoimmune diseases and cancer.

Functions of the IL17A-TH17 Pathway
IL17A could elicit crucial responses on protective immunity to fungi and bacteria infection, immune cell recruitment, and tissue repairment. IL17A also plays a key role in infection-induced immunopathology and autoimmune diseases.
Exogenous pathogens or auto-antigens generate PAMPs binding to PRRs, activate antigen-presenting cells, and release T cell-polarizing cytokines. TH17 cells, IL-17-secreting CD8⁺ (IL-17⁺CD8⁺) T cells, and IL-17-secreting γδ T (γδT17) cells recruit neutrophils and macrophages through the production of chemokines to kill exogenous pathogens and repair damaged tissues.

Treatment Strategies by Targeting the IL17A-TH17 Pathway
Various therapeutic strategies targeting the IL17A-TH17 pathway are being developed to reduce the IL17A-associated pathological effects such as inflammation, matrix destruction, and cell migration.
These reagents include inhibitors of TH17-cell generation and IL17A-IL17RA signal transduction as shown in Figure 2. Also, there are functional antibodies against IL17A-IL17RA or IL-17A/IL-17F which play antagonistic and neutralizing roles.
