IL-2Rα (p55, CD25), IL-2Rβ (p75, CD122) and IL-2Rγ (p65, CD132) are three receptors of IL-2. When IL-2 binds to them, they can produce different physiological functions. Among them, IL-2Rα, with a molecular weight of 55 kDa, is a transmembrane glycoprotein containing N-and O-linked glycosyl units that are not actively involved in receptor signaling but are specific for IL-2. Instead, it enhances receptor affinity for IL-2, leading to significant enhancement of T lymphocyte proliferation. The human IL-2Rα is also described as the Tac antigen or CD25. The deduced amino acid sequence of IL-2Rα predicts a mature protein of 251 amino acids with a signal peptide of 21 amino acids in length. As an important component of high-affinity IL-2R, the IL-2Rα chain plays an important role in the immunological specificity of clonal expansion of antigen-activated T cells. The "sushi" domain, which refers to four conserved cysteine motifs, are normally involved in protein-protein interactions, playing an important role in the presentation of IL2 (or IL15) molecules.
The sIL-2Rα has an inhibitory effect on IL-2-induced proliferation of the IL-2-dependent mouse T-cell line CTLL-2 as well as proliferation and cytotoxic activity of human PBMCs in vitro. Interestingly, the inhibitory capacity of sIL-2Rα depends on the cell density, with higher cell numbers resulting in less efficient inhibition. The most determining factor that influences sIL-2Rα function as of today's knowledge appears to be the amount of membrane-bound IL-2Rα on the target cell.

sIL-2R and IL-2Rα are present on the surface of IL-2-stimulated cells, and they are expected to be biomarkers for tumor diagnosis and prognosis, with IL-2Rα being a low-affinity receptor that is upregulated in malignant cells. sIL-2R has potential as a target for cancer therapy due to its ability to modulate the immune system and tumor microenvironment, whereas IL-2Rα has similar anticancer effects, including induction of apoptosis in multiple myeloma cells and inhibition of oncogenic pathways such as the JAK-STAT pathway. Therefore, sIL-2R and IL-2Rα have also been investigated as therapeutic targets for cancer. The clinical potential of the sIL-2Rα is not limited to its potential use as a biomarker, but might also involve its capability to modulate IL-2 function.

MAT has been shown to successfully inhibit ATL growth and prolong the survival of tumor-bearing mice.
HAT, a humanized form of MAT, has been developed. The antibody retained all the complementary determining regions from the mouse, but the Fc region was human IgG1.
