IL3RA, being the principal ligand-binding subunit of the heterodimeric receptor IL-3R, is ubiquitously expressed in IL3-responsive cells including hematopoietic stem cells, megakaryocytes, monocytes, B lymphocytes, and others. It plays a key role in mediating basic cellular behaviors such as cell proliferation, survival, and differentiation, while also participating in immune and inflammatory responses.

The Biology of IL3RA

IL3RA represents an attractive biomarker for the diagnosis and intervention of neoplastic diseases, particularly those malignancies of bone marrow and lymphatic origins. Additionally, it has been proven that the activation of IL3RA can regulate the activation and functionality of basophils or mast cells in mature immune cells.

IL3RA performs its biological functions predominantly via binding with IL-3; the assembly of IL-3/IL-3R complexes triggers the activation of the downstream networks JAK/ATAT, MAPK, and PI3K/AKT, which thereby drives hematopoietic cell development and promotes terminal differentiation of bone marrow cells.

The Pathology of IL3RA

IL3RA is overexpressed on the surface of various malignant hematological tumors including acute lymphoblastic leukemia, hairy cell leukemia, chronic granulocytic monocytic leukemia, myelodysplastic syndromes, Hodgkin's lymphoma, and more. While its pathological functionality remains not entirely elucidated, extensive screening views IL3RA as potentially correlated with poorer prognosis of tumor intervention, therefore making it an attractive diagnostic and prognostic biomarker for malignant hematological tumors.

The Therapeutic Significance of IL3RA
