Product listMUC1 (Mucin 1) is a transmembrane protein heterodimer with a heavily glycosylated extracellular domain. O-glycosylation and N-glycosylation in MUC-1 contribute to the formation of mucin. MUC1 can operate as a moisturizer, lubricant, and physical barrier in normal cells due to its structural and biochemical features. Besides, MUC1 has been demonstrated to have immunosuppressive qualities, to defend against infections, and to participate in both the oncogenic and cell signaling processes.
MUC1 as a Therapeutic Marker in Cancers
MUC1 is overexpressed in various types of cancers, including breast, lung, and pancreatic cancer, making it an attractive target for cancer therapy. Its extracellular receptor-like domain can be released into the environment and function as a decoy for mucosal pathogens detecting the environment. According to the research, MUC1 expression levels correlate substantially with colorectal cancer tumor grade. MUC1 may be found at various stages of pancreatic cancer and can accurately predict initiation and development. Furthermore, MUC1 has been linked to immune checkpoint genes, neoantigens, and specific immunotherapy prognostic indicators, implying that it might be used as an immunotherapy target and prognostic biomarker.
Function of MUC1 in Cancer
MUC1 is a well-known vital oncogene that influences a variety of cancer progression pathways through its involvement in intracellular signaling events and the control of associated biomolecules. MUC1 promotes pancreatic cancer cell invasiveness by inducing EMT. MUC1 expression is linked to VEGF expression, which promotes angiogenesis in cancer and, to a certain extent, tumor migration and invasion. MUC1 promotes cancer development and survival by regulating many pathways, including the MAPK pathway. MUC1 overexpression is also enough to cause transformation and resistance to stress-induced apoptosis.

Anti-MUC1 Antibodies Immunotherapy
- Monoclonal antibodies
MUC1-specific neutralizing antibodies are a class of antibodies that bind to MUC1 with high specificity and affinity, blocking its function and inhibiting cancer cell growth. Several neutralizing antibodies against MUC1 have been constructed and are now being tested in preclinical and clinical trials. A humanized anti-MUC1 monoclonal antibody, for example, has been proven in vitro and in vivo to inhibit the progression of advanced ovarian cancer. It works by binding to the extracellular domain of MUC1 on the surface of cancer cells, blocking its interaction with growth factor receptors and reducing cancer cell proliferation. In addition to their direct anti-tumor effects, MUC1-specific neutralizing antibodies may also enhance the activity of other cancer therapies, such as chemotherapy and immune checkpoint inhibitors.
- Bispecific antibodies
Bispecific antibodies (BsAbs) can detect two unique epitopes or antigens at the same time, enhancing immune cells' ability to target cancer cells. MUC1 has recently been explored for the design of BsAbs. According to research, a novel MUC1×CD3 BsAb increases the antitumor activity of CD3+ T-LAK cells. Besides, a MUC1×CD28 BsAb has been shown to inhibit the growth of TFK-1 cancer cells and bile duct carcinoma in SCID mice.
