NECTIN4 (Nectin Cell Adhesion Molecule 4), also known as PRR4 or PVRL4, is a well-known member of the Nectin family. Members of the Nectin family, including NECTIN1, NECTIN2, and NECTIN3, are found to be widely present in normal adult tissues, while NECTIN4 is proven to be highly expressed in embryos and placentas. More importantly, NECTIN4 is found to be aberrantly expressed in a variety of human cancers, including breast and lung cancer, revealing that NECTIN4 protein may be related to the occurrence of these cancers and may even serve as a potential target for cancer treatment.

Main Functions

As an adhesion molecule, NECTIN4 is a key factor in regulating various cell functions, such as cell motility and differentiation. In addition, studies reveal that NECTIN4 may be related to the regulation of cell size during aging. In addition to serving as a marker for multiple cancers, NECTIN4 is a compelling prognostic predictor that may aid in the treatment of ovarian, colorectal, and pancreatic cancers. It is worth noting that NECTIN4 is also a crucial regulator of adaptive immunity and angiogenesis.

NECTIN4 Pathways

  • NECTIN4-Rac1 signaling pathway

Data show that NECTIN4 regulates cell migration and lamellipodia formation via activating Rac1. Furthermore, Nectin4 is shown to be involved in the management of cell growth, angiogenesis, and proliferation in human lung adenocarcinoma cells through the Rac1 signaling pathway.

  • NECTIN4-PI3K/AKT signaling pathway

Research reveals that the PI3K/AKT signaling pathway is involved in NECTIN4-mediated growth and movement of gastric cancer cells. This indicates that NECTIN4 may serve as a potential therapeutic target for gastric cancer.

Therapeutics Targeting NECTIN4

Currently, an antibody-drug conjugate targeting NECTIN4 provides a new horizon for the treatment of metastatic urothelial cancer. In addition, monoclonal antibodies or compounds directly targeting NECTIN4 may also provide new inspiration for cancer treatment.
