Norovirus is a highly contagious, non-enveloped RNA virus belonging to the family Caliciviridae. It is the leading cause of epidemic acute gastroenteritis worldwide, affecting millions of people annually and leading to significant morbidity and economic burden. The virus is primarily transmitted through the fecal-oral route via contaminated food, water, or surfaces, and through person-to-person contact, including exposure to aerosolized vomit. Its high infectivity and environmental stability make it a formidable public health challenge. The norovirus genome contains three open reading frames (ORFs): ORF1 encodes non-structural proteins necessary for viral replication, while ORF2 and ORF3 encode the major (VP1) and minor (VP2) capsid proteins, respectively. The VP1 protein is the major structural component of the viral capsid and is essential for host cell recognition and entry. The significant genetic and antigenic diversity of noroviruses, particularly within the GII.4 genotype, poses a challenge to vaccine development, as new variants constantly emerge, escaping existing immunity.
