Neuropilin 1 (NRP1), also known as VEGF165R or CD304, is an essential member of the neuropilin family. NRP1 is a critical membrane-bound coreceptor. NRP1 contains a specific intracellular domain, a short transmembrane domain, and a large extracellular domain. This special structure allows NRP1 to participate in multiple signal transduction pathways. NRP1 binds various ligands, which provides the opportunity for it to participate in the regulation of cell survival and migration.

As a transmembrane glycoprotein, NRP1 serves as a receptor for many extracellular ligands, including vascular endothelial growth factors (for example, VEGFA), class III/IV signaling proteins, and transforming growth factor β. NRP1 has been shown to be closely connected to a variety of life activities, including cell survival, cell migration, axon guidance, and angiogenesis. Furthermore, NRP1 also plays an essential role in regulating tumor angiogenesis, tumor proliferation, and migration.
It has been reported that NRP1 activates the NF-κB pathway to participate in the regulation of keratinocyte proliferation. In addition, the VEGF/NRP1 signaling pathway formed by VEGF and NRP1 is a crucial regulator of angiogenesis.

Increasing evidence reveals that NRP1 may be related to vascularization in cancer. Additionally, abnormalities in NRP1 have been associated with the progression of several cancers, including prostate, colon, brain, breast, and lung cancers. NRP1 levels were also positively relevant to metastasis in non-small cell lung cancer. NRP1 is also closely associated with SARS-CoV-2 infection in humans. The spike protein of specific influenza viruses can bind to specific sites on NRP1 on the cell surface, thereby initiating virus entry into host cells.
Increasing evidence indicates that NRP1 is a valuable therapeutic target for anti-angiogenic therapy. Antagonists targeting NRP1 have been shown to have promising effects.
