Product listTNFRSF17, also known as B-cell maturation antigen (BCMA), is a class III transmembrane glycoprotein of approximately 20.2 kDa. TNFRSF17 cooperates with its ligand to orchestrate the maturation, survival, proliferation, and plasma cell differentiation of B cells.
Under physiological conditions, the expression of TNFRSF17 is primarily concentrated on the surface of plasma cells and plasmablasts. It has been demonstrated in associated research that TNFRSF17 is extensively expressed on the surface of multiple myeloma (MM) cells.

TNFRSF17 Biology
Four natural splice variants of TNFRSF17 have been identified to date, each with different membrane anchoring capabilities, intracellular structure, and signal and receptor-binding affinity. The ligands of TNFRSF17 include BAFF and APRIL.
Comparatively, APRIL exhibits higher specificity and binding affinity to BCMA, and it is associated with a greater number of downstream pathophysiological processes. Additionally, TNFRSF17 can be directly shed by gamma-secretase activity to form soluble sBCMA.

TNFRSF17 Signaling Pathway and MM Cells
TNFRSF17 is expressed on nearly all MM cell surfaces and participates in their pathological progression. Upon binding to its ligand, TNFRSF17 can activate the NF-κβ, PI3K-PKB/Akt, and RAS/MAPK signaling pathways in MM cells. These signals can modulate cell cycle checkpoints, stimulate proliferation, and upregulate anti-apoptotic proteins, angiogenic factors, cell adhesion molecules, and immunosuppressive molecules.

TNFRSF17 Therapeutics
The overexpression and activation of TNFRSF17 have significant correlations with the progress of human MM and it is upregulated in malignant PC. Consequently, it is considered an effective biomarker and a promising target for MM intervention.
The currently developed TNFRSF17-targeted intervention strategies include bispecific T cell engagers, chimeric antigen receptor T cells, and antibody-drug conjugates. These strategies have demonstrated efficacy in preclinical models and show potential in clinical trials.
