CD274 molecule (programmed death ligand 1), also known as PD-L1, PDCD1L1, and B7H1, encodes the ligand for immune inhibitory receptor PDCD1/PD-1. CD274 is expressed on the surface of the cells, such as T cells, B cells, and various types of tumor cells. It influences the regulation of the immune system in the aspects of autoimmunity and tumor immune escape.
Function
Functions for CD274 include:
- Inhibits T-cell activation and cytokine production.
- Inactivates cytotoxic T lymphocytes and facilitates tumor survival.
- Maintains the homeostasis of the immune response and prevents autoimmunity.
Signal Pathway
The CD274 inhibitory pathway is being widely exploited to normalize the anti-tumor immune response for cancer immunotherapy. The hyperactive oncogenic pathways that are listed below play a vital role in CD274 expression.
- Epidermal Growth Factor Receptor (EGFR) pathway.
- Mitogen-activated protein kinase (MAPK) pathway.
- PTEN/PI3K-AKT pathway.
- JAK-STAT pathway.
- NF-κB pathway.
Drug Development strategy
- Antibodies targeting PDL1 and acting as immune checkpoint inhibitors can disrupt the interaction between PD1 and PDL1 and induce a durable anti-tumor response.
- PDL1 antibody-drug conjugate (ADC) may serve for chemo-guided immune therapy and image-guided tumor diagnosis. As shown in figure 1, PDL1-Dox could specifically target the tumor cells and destroy tumor stromal components, resulting in improved T-cell immunity.

- Regulation of PD-L1 expression may improve the current immune checkpoint inhibitor therapy. As shown in figure 2, a CDK4/6 inhibitor together with antibodies that block PD-L1 could overcome immune evasion. Metformin together with CTLA-4 antibodies could exhibit a synergistic antitumor activity.
