Inflammatory Pathway of IL1B
The pro-inflammatory cytokine IL1B gets released and activated by the NLRP3-induced pyroptosis process of a subtype of immune cells expressing IL1B. IL1B activates the signaling pathways promoting tumor invasion and metastasis.

Immune Functions of IL1B
Overexpression of IL1B by cancer cells leads to the accumulation of MDSCs and macrophages, inhibits the function of NK cells, activates neutrophils, and triggers the release of pro-tumor cytokines. IL1B in the tumor microenvironment promotes a series of pro-tumor immune responses. As shown in Figure 2, IL1B released by dendritic cells could initiate T-cell responses and enhance the inhibition of tumor cell growth.

Clinical Significance of IL1B
IL1B plays a pivotal role in cancer genesis and development. Many therapeutic approaches with different kinds of biological agents targeting IL-1 are summarized here (Fig.3). Fig.3a shows the agents blocking IL-1, including anti-IL1B antibody, sIL-1RI, and IL-1 trap. Fig.3b depicts the agents competing with IL-1R and blocking IL-1R, such as IL-1Ra and anti-ILRI antibodies. And the caspase 1 inhibitors reduce the production of IL1B, see Fig.3c.
