Product listTriggering receptor expressed on myeloid cells 2 (TREM2) has three isoforms as results of the pre-mRNA alternative splicing. Isoform 1 is a membrane-bound protein while isoforms 2 and 3 are secreted outside the cell membrane. Defects of TREM2 may cause some diseases including microglia inflammation, neurodegenerative diseases, and Alzheimer's disease (AD).

Structure and Signaling Pathways of TREM2
TREM2 ligands contain ApoE, HDL, LDL, and Aβ. The interaction between TREM2 and its ligands can induce the association of TREM2 to DAP12 or DAP10. Tyrosine phosphorylated DAP12/10 recruits SYK which initiates various activation of signaling pathways (see in Fig.1).

Functions of TREM2
Through the amplification of TREM2-ligand signals, TREM2 remodels the cytoskeleton and influences cell functions such as cell survival and metabolism. Here are four models showing the function of TREM2 in AD.
- TREM2 promotes cell phagocytosis and enhances the removal of Aβ (Fig.2a).
- TREM2 may sustain the long-term survival and the activation state of microglia during AD (Fig.2b).
- TREM2 helps recruit peripheral monocytes through the blood-brain barrier to reduce the Aβ plaque burden (Fig.2c).
- TREM2 initiates a signal suppressing the aging risk factors to delay aging and sustain the survival of microglia (Fig.2d).
