Product listGPC3 (Glypican 3) is a membrane-associated proteoglycan that is found on the surface of cells in the human body. It plays a role in many kinds of biological activities, including cell growth and division, as well as the development of various tissues and organs. GPC3 is overexpressed in hepatocellular carcinoma (HCC), despite being expressed seldom or not at all in normal liver tissues, making it an ideal diagnostic and therapy target for HCC.
Biological Function of GPC3
Mutations in the GPC3 gene have also been associated with Simpson-Golabi-Behmel syndrome (SGBS), a rare genetic disorder that affects many parts of the body. GPC3 is an extracellular matrix (ECM) component that can interact with a range of growth factors, chemokines, and cytokines, consequently maintaining the concentration of extracellular ligands and increasing their binding to their associated receptors. Besides, GPC3 participates in multiple signaling pathways in HCC. GPC3 has been shown in studies to help in the stimulation of WNT signaling by boosting the development of membrane surface complexes in liver cancer. C-myc has also been found to be associated with the oncogenic stimulation of GPC3 in HCC, regulating cell growth, proliferation, and differentiation.

GPC3-Targeted Antibodies Therapy for HCC
- Monoclonal antibodies
GPC3-specific neutralizing antibodies are a class of antibodies that bind to GPC3 with high specificity and affinity, blocking its function and inhibiting cancer cell growth. Several neutralizing antibodies against GPC3 have been constructed and are currently being tested in preclinical and clinical trials. One such example is a humanized monoclonal antibody that has been shown to inhibit the growth of GPC3-positive HCC cells in vitro and in vivo. It works by binding to GPC3 on the surface of cancer cells, blocking its interaction with Wnt signaling pathway proteins, and reducing cancer cell proliferation. In hepatoma cells, researchers have discovered a human monoclonal antibody against the GPC3 HS chain that inhibits HGF/c-Met pathway-mediated migration and motility. In addition to their direct anti-tumor effects, anti-GPC3 antibodies may also enhance the activity of other cancer therapies, such as chemotherapy and immune checkpoint inhibitors.
- Bispecific antibodies
In the future, anti-GPC3 bispecific antibodies might be a potential method for HCC therapy. A T cell-redirecting antibody (TRAB) with a humanized IgG structure and a shared light chain may bind to both GPC3 and CD3, boosting cytotoxicity via T cell effectors. A bispecific antibody that targets both GPC3 and CD47 has proved efficient in inhibiting tumor growth by binding to both antigens.
