Product listLymphotoxin alpha (LTA) is also known as TNFSF1 or tumor necrosis factor-beta (TNFB). Produced by lymphocytes, LTA is a cytokine member belonging to the tumor necrosis factor family. The LTA homotrimer protein is secreted while the LTA heterotrimer protein is membrane-bounded. LTA is associated with autoimmune diseases such as psoriatic arthritis and leprosy.

Functions of the LTA Signaling
LTA has a high cancer therapeutic potential with the pro-tumor mechanisms of LTA in malignant Hodgkin tumor cells. LTA has the following functions:
- With the mutations or loss of A20, LTA and its receptor TNFRSF14 activated by NF-κB drive a constitutive NF-κB activation in a feedback loop, and promote the production of cytokines, chemokines, and receptors.
- LTA regulates a constitutive activation of JAK2/STAT6. Phosphorylated STAT6 enhances the expression of adhesion molecules and immune checkpoint ligands such as PD-L1/PD-L2, which promote tumor invasion and the development of tumor drug resistance.

Treatment Strategy for LTA
Anti-tumor strategies based on the blockade of the LTA axis have been developed for the treatment of LTA pathological diseases and all kinds of cancers such as lymphoma. The strategies disrupting the interaction of LTA signaling may be explored.
- Bispecific antibodies and ADCs target and block the LTA axis.
- Immune checkpoint inhibitors against PD-L1/PD-L2 may generate a synergistic therapeutic effect.
