Summary

Description This product is a monoclonal antibody which can be used to selectively bind to proteolytically active, high molecular weight ADAM10 protease. This antibody recognises a specific conformation of the ADAM10 substrate binding domain dependent on CxxC bonding. The mAb is capable of distinguishing tumour cells from normal or non-malignant cells.

Application Notes Antibody clone 8C7 clearly inhibits signalling by Notch, and also Eph receptors, however the antibody does not interact directly with residues identified as contributing to substrate binding. The mAb recognises a specific conformation of the ADAM10 substrate binding domain dependent on CxxC bonding. 8C7 binding is blocked by mutation of the CxxC motif, and is altered by modulating the redox environment. mAb 8C7-immunoprecipitates of ADAM10 showed highly significant enrichment of protease activity. Also, oxidative conditions, known to enhance ADAM activity, favoured 8C7 binding.
