Description | The antibody 10-40 is a human mAb specific for SARS-CoV-2 Spike S1 protein. It is a human monoclonal antibody, that neutralized or bound all sarbecoviruses tested in vitro and protected against SARS-CoV-2 and SARS-CoV in vivo. Comparative studies with other receptor-binding domain (RBD)-directed antibodies showed the antibody to have the greatest breadth against sarbecoviruses, suggesting that it is a promising agent for pandemic preparedness. |

Application Notes | 50 ng per well of antigen (S trimer or RBD protein) was coated onto ELISA plates at 4°C overnight. The ELISA plates were then blocked with 300 μL blocking buffer consisting of PBS with 1% BSA and 10% bovine calf serum at 37°C for 2 hours. Afterwards, 100 μL of serially diluted antibodies were added and then incubated at 37°C for 1 hour. Next, 100 μL of 10,000-fold diluted Peroxidase AffiniPure goat anti-human IgG Fcγ fragment specific antibody was added into each well and incubated for an other 1 hour at 37°C. The plates were washed between each step with PBS-T. Finally, TMB substrate was added and incubated before the reaction was stopped using 1 M sulfuric acid. Absorbance was measured at 450 nm. |
