Application Notes | ELISA Assay of murine 5E3 (mu5E3) mAb An ELISA was used to test the binding of mu5E3 against cSNK-BSA peptide. The ELISA plate was coated with antigen (Ag), 1 μg/well of cSNK-BSA. The wells were blocked with 5% skim milk then probed with serially diluted the mAbs (0.01 μg/mL to 5 μg/mL) and binding was detected with a commercial goat anti-mouse HRP conjugate antibody. The plate was read at 450 nm. Western Blot Analysis of murine 5E3 (mu5E3) mAb A 4-12% gradient SDS-PAGE gel was run with cSNK-BSA. The gel was then transferred to a nitrocellulose membrane. The next day the mAbs were diluted to 2 μg/ml to 5 μg/ml depending on the antibody, and used to probe the membrane at room temperature (RT). The membranes were then probed with anti-mouse IgG-HRP. |
