| Application Notes | The binding affinity of the antibody was measured at 25, 50, or 100 nM against the immobilized peptide of GPPPPIDNGDITSFP using SPR in a BIAcore instrument. This analysis revealed an off-rate (kd) of 5.56×10-7 s−1, an on-rate (ka) of 2.26×105 M−1 s−1 and an affinity (KD) of 2.46 nM. Complement-Dependent Cytotoxicity (CDC) assay A549 lung cancer cells were incubated with CFH antibody 7968 along with normal human serum (NHS) as a source of complement. The antibody was tested at 60 or 120 μg/mL and measured cytotoxicity by an LDH-release ELISA. The results showed that mAb 7968 caused a statistically significant increase in CDC at 60 μg/mL and 120 μg/mL. |
