Description | This is a neutralizing antibody clone 8A10 specific for CVB1 capsid protein, derived from mouse. 8A10 binds to an epitope closer to the viral canyon area and has high binding affinity toward the CVB1. It displays a high neutralizing activity against CVB1. The antibody has been determined by Enzyme-linked immunosorbent assay, Immunofluorescence, Bio-layer interferometry and Neutralization assay. |
Clonality | Monoclonal |
Host Species | Mouse |
Target Species | Coxsackievirus B1 (CVB1) |
Epitope | 8A10 binds to an epitope closer to the viral canyon area. The epitope of 8A10 mainly consists of residues in the VP1 HIloop (Q254, E256, K257, P265, I266, and G267), the VP2 EFloop (aa 136-138 and aa 159-163) and the VP3 N terminus (aa 232-238). |
Neutralization Mechanism | 8A10 binds to an epitope closer to the viral canyon area and inhibit CAR binding to the virus. |
Neutralization Breath | The middle dosage (0.1 mg/kg) of 8A10 offered 100% protection against lethal challenge. |

Application | ELISA; SPR; IF; Neut |
Application Notes | ELISA: mAb at various concentrations was first incubated at 37°C for 30 min, then combined with the soluble CAR (2 mg/mL) and further incubated at 37°C for 30 min. Surface plasmon resonance (SPR): MAb was directly immobilized onto a Protein G biosensor chip (Cytiva) using a PBS running buffer. Immunofluorescence (IF): After blocking with 2% bovine serum albumin (BSA) diluted in PBS, cells were incubated with mAbs (5 μg/mL) at 37°C for 1 h. Neutralization assay (Neut): MAb was 2-fold serially diluted and incubated with an equal volume of CVB1 at 37°C for 1 h. |
