Application Notes | The IC50 of BG4-25 is 0.004 μg/mL. MaxiSorp 384-well micro-plates were coated with 50 μl/well of 2,500 ng/ml of SARS-CoV-2 RBD in coating buffer overnight at 4°C. Plates were then washed 3 times with 100 μl/well of wash buffer using a BioTek 406 plate washer. Plates were blocked by adding 100 μl/well of blocking buffer for 30 min at room temperature. Plates were then washed as described above. 50 μL of 1:100 diluted serum samples in dilution buffer were added to the wells and incubated for 30 min at 37°C. Plates were then washed 7 times as described above. 50 μl/well of 1:25,000 diluted detection Ab solution was added to the wells and incubated for 30 min at room temperature. Plates were then washed 7 times as described above. 40 μl/well of Pierce TMB peroxidase substrate was then added to the wells and incubated at room temperature for 3 min or 5 min. The reaction was then stopped by adding 40 μl/well of stop solution to each well. The OD was read after 15 min at 450 nm and 570 nm on a BioTek Synergy HT using Gen5 software. |
