Summary

Description The mouse antibody TyTx4 recognizes the typhoid toxin. TyTx4 binds to PltB subunits and its epitope overlaps with glycan receptor-binding sites on PltB subunits. It neutralizes the increased dose of typhoid toxin effectively and it almost completely inhibits typhoid toxin binding to brain endothelial cells in vivo assay.
Clonality Monoclonal
Host Species Mouse
Target Species Salmonella typhi
Immunogen Recombinant typhoid toxoid
Epitope Its epitope overlaps with glycan receptor-binding sites on PltB subunits.
Neutralization Mechanism TyTx4 inhibits typhoid toxin binding to cell surface glycan receptors and results in neutralization of typhoid toxin-mediated cellular toxicity.
Affinity 0.028 nM
Isotype Mouse IgG2a

Applications

Application ELISA; WB; SPR; IF; FC; Neut
Application Notes This antibody was tested using ELISA, Western Blot (WB), Surface plasmon resonance (SPR), Immunofluorescence (IF), Flow cytometry (FC) and Neutralization assay.
Surface plasmon resonance (SPR): Pilot experiments to optimize experimental parameters were then performed using 10-fold serial dilutions of each MAb injected at a flow rate of 50 μL/min with a contact time of 120 sec and dissociation time of 600 sec.
Immunofluorescence (IF): The slides were stained with indicated antibodies for 2 hrs at room temperature, counterstained with DAPI, mounted using ProLong antifade mounting solution, and viewed.
Neutralization assay (Neut): Infected cells were then incubated in complete cell culture medium containing 10 μg/mL gentamicin for 72 hours in the presence and absence of 10 nM MAbs.
