Application Notes To determine specificity of recombinantly produced mAbs, 96 half area well-plates were coated over-night at 4°C with of SARS-CoV-2 S, NTD or RBD proteins prepared at 1 μg/mL, 2 μg/mL and 5 μg/mL in PBS pH 7.2, respectively. Plates were then blocked with PBS-1% BSA and subsequently incubated with mAbs serial dilutions for 1 h at room temperature. After 2 washing steps with PBS-T goat anti-human IgG secondary antibody was added and incubated for 1 h at room temperature. Plates were then washed again with PBS-T and 4-NitroPhenyl phosphate substrate added. After 30 min incubation, absorbance at 405 nm was measured by a plate reader and data plotted using Prism GraphPad.
