Adeno-associated virus (AAV) is a compact, non-enveloped virus commonly employed as a gene delivery vector owing to its minimal immune reactivity. Its protein coat, or capsid, is assembled from VP1, VP2, and VP3 subunits, which play a key role in defining the virus's tropism—the capacity to selectively enter and transduce particular cell types. The complex architecture of the capsid and minor variations among VP proteins across serotypes contribute to AAV's versatility in diverse gene therapy uses. Each of the more than a dozen identified AAV serotypes (such as AAV1, AAV2, AAV5, AAV8, and AAV9) exhibits unique tissue targeting preferences, rendering them well-suited for transporting genetic payloads to designated tissues like the liver, muscle, or central nervous system. Despite being non-pathogenic, a major hurdle in gene therapy is that numerous people carry prior immunity to several AAV serotypes due to natural infection. This may result in vector clearance by the host's immune defenses, thereby diminishing treatment effectiveness. These factors underscore the necessity for accurate AAV profiling and neutralization antibody assessment prior to administering gene-based therapies.

A: These products are primarily used for the detection, characterization, and quantification of AAV viral vectors. They are invaluable for quality control assays in gene therapy manufacturing, for studying the biodistribution of AAV vectors in preclinical models, and for developing assays to screen for pre-existing neutralizing antibodies in patient populations.
A: The choice of antibody depends on the AAV serotype you are using in your research. Our product line offers antibodies specific to a wide range of common serotypes, such as AAV1, AAV2, AAV5, AAV8, and AAV9. If you are unsure, our scientific support team can help you select the most suitable product for your specific application and AAV vector.
A: Our antibodies are primarily validated for in vitro applications such as ELISA, Western blotting, and immunofluorescence. While some researchers have successfully used similar antibodies for in vivo studies, such as blocking AAV transduction, this requires careful consideration of the specific antibody's characteristics and host organism. We always recommend contacting our team to discuss your specific in vivo needs.
A: For clinical applications, our antibodies can be used as key reagents in neutralizing antibody assays. These assays help determine if a patient has pre-existing antibodies that could inactivate the AAV vector, which is a critical step in patient stratification for gene therapy trials.
