CD19, a 95 kD Type I transmembrane protein, is abundantly expressed on the surface of B lymphocytes. Serving as a biomarker for both normal and tumourous B cells as well as follicular dendritic cells, CD19 regulates B-cell receptor-induced signals or intrinsic signals to participate in the establishment of B cell signaling threshold, thereby modulating central responses of B lymphocytes. As CD19 expression is upregulated in tumor B cells, it is considered to be a potential interventional strategy for B-cell lymphoma or leukemia.

Identified as the B4 antigen of human B lymphocytes, CD19 is regarded as one of the most reliable surface markers of B cells as well as a key co-receptor for BCR signal transduction.
CD19 not only acts as a bridging protein to recruit cytoplasmic signaling proteins to the membrane surface, but also functions as a signaling subunit of the CD19/CD21 complex when coupled with BCR. Through BCR-associated or independent signal transduction processes, CD19 appropriately lowers the threshold for antigen receptor-dependent stimulation, thereby activating B cells with fewer receptors involved.

Dysfunctions of CD19 can lead to low responsiveness of B cells to receptor stimulation signals accompanied by weaker T-cell dependent humoral responses, causing the failure of humoral immune responses. This discovery has sparked significant interest in targeting CD19 to intervene in various autoimmune diseases.
Furthermore, research has shown that CD19 is highly expressed on the surface of most B-cell malignancies. Consequently, targeted anti-CD19 monoclonal antibodies, Chimeric Antigen Receptor (CAR) T-cell immunotherapy, and bispecific T-cell engager (BiT) antibodies have been widely developed and used for interventions in diseases like lymphoma.
