CD96 (Cluster of Differentiation 96), also known as T-Cell Surface Protein Tactile or TACTILE, is a pivotal receptor protein. As an immunoglobulin superfamily transmembrane glycoprotein, CD96 is found to be expressed in both mice and humans. In humans, CD96 is mainly expressed in natural killer cells (NK cells), B cells, and hematopoietic stem cells. In mice, CD96 is present in NK cells and NKT cells. Most importantly, the expression level of CD96 in mice is higher than that in humans. Curiously, CD96 is not found to be expressed in other immune cells.

As a co-stimulatory receptor, CD96 contributes to the enhancement of CD8+ T cell activation and effector responses. It has been reported that CD96 is involved in regulating NK cell adhesion to target cells. CD96 also plays a vital role in inhibiting NK cell-induced cytokine responses. CD96 may also be associated with antigen presentation. In addition, CD96 is an important hub in the immune response, collaborating with other checkpoint members. Therefore, CD96 is gradually emerging as an attractive candidate for immunotherapy.
Abnormal expression of CD96 is closely linked to various diseases, including myelodysplastic syndromes and acute myeloid leukemia. Currently, accumulating data indicate that CD96 is differentially expressed between normal tissues and cancer tissues. Notably, data show a significant correlation between immune evasion and CD96 in hepatocellular carcinoma tissues, suggesting that CD96 may play a role in tumor cell evasion.
CD96 is reported to signal mainly through the canonical MEK-ERK pathway. CD96-induced phosphorylation of these kinases is comparable to the co-stimulatory effect of CD226. It has been reported that blocking CD96 expression on CD8+ T cells by antibodies impairs the expression of transcription factors and proinflammatory cytokines associated with CD8+ T cell activation. This result suggests that CD96 signaling may be involved in CD8+ T cell activation and differentiation.
