Introduction of E. coli O157 H7

Escherichia coli O157:H7 is a highly virulent serotype of enterohemorrhagic E. coli (EHEC) and a major global foodborne pathogen, identified by its O157 somatic and H7 flagellar antigens. Transmitted mainly through contaminated food (like undercooked beef, unpasteurized milk, and produce), its low infectious dose necessitates robust, rapid detection. Pathogenicity centers on two factors: the production of potent Shiga toxins (Stx1 and Stx2), which damage the vascular endothelium, especially in the kidneys; and the ability to create "attaching and effacing" (A/E) lesions on the intestinal epithelium. The resulting clinical syndrome, Hemorrhagic Colitis, can progress to the life-threatening Hemolytic Uremic Syndrome (HUS), which causes acute renal failure and neurological damage. Diagnostic accuracy based on both O157 antigens and Shiga toxins is vital for patient management and epidemiological tracking.

Antibodies Against E. coli O157 H7

Creative Biolabs' Neutra™ Antibodies target the defining structures and virulence factors of E. coli O157:H7, offering researchers and assay developers unparalleled precision across various platforms.

Targeting Mechanisms:

  • O157 Antigen Detection: Monoclonal antibodies targeting the O157 lipopolysaccharide (LPS) identify the serotype, serving as the primary capture agent for high-throughput screening of bacterial cultures or swabs.
  • H7 Flagellar Antigen Detection: Antibodies against the H7 flagellin protein provide secondary serotype confirmation, often used in confirmatory assays to enhance specificity.
  • Shiga Toxin Detection (Stx1/Stx2): Specific antibodies enable neutralization assays in research and direct diagnostic detection of free toxins in stool or culture supernatants, correlating directly to severe disease risk.

Applications in Diagnostics and Research:

These reagents span the entire detection workflow: from sample preparation (IMS) to final confirmation (ELISA/Western Blot). By targeting the cell surface, flagella, and secreted toxins, our antibody portfolio supports comprehensive, layered detection systems adhering to stringent food safety and clinical laboratory requirements.

Why Choose Us?

Choosing Creative Biolabs ensures partnering with a specialist committed to maximum assay performance. Our E. coli O157 H7 specific antibodies are developed rigorously to solve real-world application challenges.

  • Exceptional Specificity: Antibodies are cross-screened against related E. coli strains to minimize off-target binding and reduce false positives in complex samples.
  • High Affinity and Sensitivity: Reagents are optimized for high binding affinity, enabling lower limits of detection (LOD) crucial for early contamination identification in food.
  • Versatile Platform Compatibility: Neutra™ antibodies are validated for multiple platforms, including ELISA, Lateral Flow, and IMS, ensuring seamless integration into new or existing kit designs.
  • Validated Performance Data: Published data confirms superior performance and stability, particularly in challenging applications like environmental and high-fat matrices.

We ensure every reagent meets strict quality standards, providing the consistency necessary for regulated environments and reproducible experiments.

FAQs

Q: What do high-specificity antibodies recognize?

A: They recognize defining surface antigens (LPS/O157 serotype and H7 flagellin) and secreted virulence factors (Shiga toxins Stx1 and Stx2), allowing for a multi-faceted approach to detection and characterization.

Q: Where are these antibodies best used for rapid detection?

A: High-affinity reagents are optimally suited for rapid, high-throughput Enzyme-Linked Immunosorbent Assays (ELISA) and Lateral Flow Assays (LFA) for time-sensitive applications like food safety surveillance.

Q: How can false-positives be minimized?

A: Minimize false positives by using cross-reactivity validated antibodies and incorporating multiple detection steps, such as a two-step antibody system or Immunomagnetic Separation (IMS) for sample purification.

Q: Why use antibody detection over culture methods?

A: Immunoassays offer significant speed (results in hours) and can detect non-viable bacteria or free toxins, which is crucial for rapid public health decision-making, complementing traditional culture methods.

Q: Can these reagents measure bacterial load or toxin concentration?

A: Yes. When incorporated into quantitative assays like a standard sandwich ELISA with a standardized curve, the reagents can accurately measure the concentration of target antigens or toxins.
