HPeVs VP1 Specific Neutra™ Antibody Products

Product list

Our HPeVs VP1 Specific Neutra™ Antibody Products help you accelerate drug discovery and enhance diagnostic precision through high-affinity monoclonal technology and structurally-validated neutralizing scaffolds. By targeting the highly variable and immunodominant VP1 capsid protein, Creative Biolabs provides the specificity required to distinguish between human parechovirus serotypes and neutralize viral entry.

Introduction of HPeVs VP1

Human Parechoviruses (HPeVs) are non-enveloped, positive-sense single-stranded RNA viruses belonging to the Picornaviridae family. While they share similarities with Enteroviruses, HPeVs are distinct in their capsid architecture, composed of only three structural proteins: VP0, VP1, and VP3. Unlike other picornaviruses, the VP0 precursor remains uncleaved into VP2 and VP4, creating a unique surface topography.

Structure and Function

The VP1 protein is the most external and immunologically significant component of the HPeV capsid. It often contains a C-terminal Arginine-Glycine-Aspartate (RGD) motif, which acts as the primary ligand for host integrin receptors. VP1 forms the five-fold icosahedral vertices of the virus, serving as the "molecular key" for entry.

Clinical Impact and Disease

HPeVs are significant pathogens in pediatric medicine. While HPeV1 typically causes mild respiratory or gastrointestinal distress, HPeV3 is frequently associated with severe, life-threatening conditions in infants under three months of age, including:

  • Neonatal Sepsis-like Syndrome: Characterized by high fever, irritability, and erythematous rash.
  • Central Nervous System (CNS) Infections: Including meningitis and encephalitis, leading to long-term neurodevelopmental sequelae.
  • Cardiomyopathy: Acute heart failure in severe neonatal cases.

Signaling and Pathogenesis

Upon entry, HPeVs trigger innate immune responses primarily through Toll-like receptors (TLRs), particularly TLR3 and TLR7/8, which sense double-stranded RNA intermediates during replication. However, the absence of maternal neutralizing antibodies is the primary risk factor for severe disease in neonates, making the development of exogenous neutralizing antibodies a clinical priority.

Applications of Anti-HPeVs VP1 Neutralizing Antibodies

The versatility of anti-VP1 neutralizing antibodies makes them indispensable in both the laboratory and the clinic:

  • Therapeutic Development: As HPeV3 lacks an RGD motif and utilizes unknown receptors, neutralizing antibodies that target quaternary epitopes are the gold standard for developing passive immunization strategies.
  • Diagnostic Assays: High-specificity mAbs are used in capture ELISA and lateral flow assays to differentiate between HPeV serotypes in clinical stool or CSF samples.
  • Vaccine Research: Used as positive controls in Microneutralization Tests (MNT) to evaluate the potency of experimental HPeV vaccine candidates.
  • Epitope Mapping: Employing Cryo-EM and peptide scanning to identify "vulnerable" sites on the capsid surface for next-generation drug design.

Our Anti-HPeVs VP1 Neutralizing Antibodies

Creative Biolabs offers a comprehensive portfolio of HPeVs VP1 Specific Neutra™ antibodies, meticulously validated for performance:

  • Serotype-Specific Monoclonals: High-affinity antibodies for HPeV1 and HPeV3 with no cross-reactivity to common Enteroviruses (e.g., EV-A71, CVA16).
  • Broad-Spectrum Cross-Neutralizers: Engineered antibodies targeting conserved residues in the VP1-VP3 interface capable of neutralizing multiple HPeV genotypes.
  • Custom Conjugations: Available with HRP, Biotin, or Fluorophores for streamlined integration into your existing workflows.

Each lot undergoes rigorous quality control, including Plaque Reduction Neutralization Tests (PRNT) and TCID50 inhibition assays, ensuring that the product you receive translates directly to biological results.

Creative Biolabs is committed to providing the global scientific community with high-performance HPeVs VP1 Specific Neutra™ Antibody Products.
