Product list

Are you currently facing challenges in antibody development for respiratory viruses, such as cross-reactivity issues or low neutralization efficiency in minor group rhinoviruses? Our HRV2 VP2 Specific Neutra™ Antibody Products help you develop highly specific therapeutics and diagnostic assays for Human Rhinovirus 2 through our advanced high-affinity monoclonal platform and precision epitope targeting of the VP2 capsid protein.

Introduction of HRV2 VP2

Human Rhinovirus 2 (HRV2) is a member of the Enterovirus genus within the Picornaviridae family. As a "minor group" rhinovirus, HRV2 is distinguished by its use of the low-density lipoprotein receptor (LDLR) family for cell attachment and entry, rather than the ICAM-1 receptor used by major group serotypes.

Structure and Function of VP2

The HRV2 virion is a non-enveloped, icosahedral capsid composed of 60 protomers, each containing four structural proteins: VP1, VP2, VP3, and VP4. Viral Protein 2 (VP2), alongside VP1 and VP3, forms the external surface of the capsid. A critical feature of VP2 is the βE-βF loop, which contains a highly immunogenic linear epitope (antigenic site B). Literature indicates that this loop undergoes significant conformational adjustment during the uncoating process, where the N-terminus of VP2 moves inward to facilitate the release of the (+)ssRNA genome into the host cell cytoplasm.

Clinical Impact and Signaling

HRV2 is a primary causative agent of the common cold but is increasingly recognized for its role in severe respiratory complications. Clinical data links HRV2 infection to the exacerbation of chronic obstructive pulmonary disease (COPD) and asthma. Following receptor binding, the virus is internalized via clathrin-dependent endocytosis. The acidic environment of the endosome triggers the "A-particle" transition, a key step where VP2 reorganization is vital for membrane pore formation and successful viral replication.

Applications of Anti-HRV2 VP2 Neutralizing Antibodies

Neutralizing antibodies (NAbs) targeting the VP2 protein are indispensable in both therapeutic research and diagnostic applications due to their ability to physically obstruct the viral life cycle.

  • Antiviral Screening Platforms: NAbs are used to validate high-throughput screening (HTS) systems, ensuring that potential inhibitors effectively compete for the same critical binding sites on the VP2 loop.
  • In Vitro Neutralization Assays (PNA): Our antibodies are utilized in microneutralization tests to inhibit cytopathic effects (CPE) in HeLa or Ohio-HeLa cell lines, providing a gold standard for measuring viral infectivity.
  • Mechanistic Research: Researchers apply anti-VP2 NAbs to study "capsid breathing"—the dynamic exposure of internal epitopes—which is essential for understanding how minor group rhinoviruses evade the host immune system.
  • Vaccine Immunogenicity Testing: In vaccine development, these antibodies serve as reference reagents to evaluate the breadth and potency of the immune response elicited by novel capsid-based antigens.
