Are you currently facing challenges in serotype-specific detection or the risk of antibody-dependent enhancement (ADE) in your dengue research? Our HSV2 gD Specific Neutra™ Antibody Products: Shielding Your Research Against Viral Entry! Are you currently facing high background noise in viral assays, low neutralization potency in candidate screening, or challenges in mapping the complex fusion machinery of Herpes Simplex Virus? Our HSV2 gD Specific Neutra™ Antibody Products help you achieve definitive viral inhibition, precise epitope characterization, and accelerated therapeutic benchmarking through high-affinity binding to glycoprotein D, the master key of HSV-2 cellular entry.
Herpes Simplex Virus Type 2 (HSV-2) remains a global health burden, primarily causing genital herpes and significantly increasing the risk of HIV-1 acquisition. Central to its pathogenesis is Glycoprotein D (gD), a structural component of the viral envelope that serves as the primary receptor-binding protein.
Structure and Function
The gD molecule consists of an N-terminal signaling domain and a C-terminal extension that wraps around the core. In its "closed" state, the C-terminus obscures the N-terminal loop. Upon binding to cellular receptors, specifically Herpesvirus Entry Mediator (HVEM) or Nectin-1, gD undergoes a dramatic conformational change. This "unmasking" event triggers a signal transduction cascade to the gH/gL complex, which eventually activates the fusogen gB to merge the viral and host membranes.
Structure and Function
The gD is the primary attachment protein that determines host cell tropism. It binds to receptors such as HVEM, Nectin-1, or 3-O-sulfated heparan sulfate. Upon binding, gD undergoes a conformational change that triggers the activation of gH/gL, initiating membrane fusion. This interaction also activates the NF-κB and PI3K/Akt pathways, promoting cell survival and inflammatory responses. Furthermore, gD binding to HVEM can interfere with BTLA signaling, effectively modulating the host's immune response to facilitate viral persistence.
Clinical Impact and Disease
The interaction between gD and HVEM is not merely structural; literature suggests it may modulate the host's innate immune response by dampening local chemokine production. Clinically, gD is the most immunogenic protein on the virion, making it the primary target for neutralizing antibodies. Its role in viral "surfing" along filopodia and subsequent cell-to-cell spread makes it a focal point for preventing both primary infection and the reactivation of latent virus in sensory neurons.
