Interleukin 21 receptor (IL21R), also known as CD360 or IL-21R, is a member of type I cytokine receptors. IL21R is expressed in diverse immune cells, such as macrophages, T cells, and natural killer (NK) cells. IL21R plays a crucial role in transducing the growth-promoting signal of IL21 in the proliferation of T cells, B cells as well as NK cells. Anti-IL21R neutralizing antibodies capable of inhibiting IL21R activity have been developed for the treatment of inflammatory disease.

IL21R Signaling Pathways

The ligand binding of IL21R is closely related to the activation of multiple downstream signaling molecules, such as Janus Kinase 1 (JAK1), JAK3, and signal transducer and activator of transcription 1 (STAT1). IL21R-mediated signaling has an essential impact on T cell activation, differentiation as well as proliferation. Furthermore, this signaling pathway is closely linked to changes in chemokine and cytokine production and effector function. Studies have shown that IL21/IL21R is also involved in the regulation of neutrophil-mediated pathological immune responses during chlamydial respiratory infections.

Related Diseases

Some evidence has revealed that IL21/IL21R signaling plays a crucial role in some idiopathic autoimmune diseases. For example, IL21R signaling is critical for the induction of autoimmune encephalomyelitis. In addition, data show that serum IL21 levels are elevated in inflammatory bowel disease (IBD) patients, revealing that IL21/IL21R signaling may be linked to the pathogenesis of IBD. Other diseases in which IL21/IL21R are implicated include chronic diarrhea, tinea corporis, cold sores, mycobacterial infections, and hypogammaglobulinemia. Detailed analysis of these individual immune cells has revealed that therapeutic manipulation of IL21/IL21R activity offers promising prospects for immunotherapy of these inflammatory diseases.
