Listeria monocytogenes (L. monocytogenes) is a ubiquitous Gram-positive bacterium that causes listeriosis, a serious foodborne illness with high mortality, especially in vulnerable populations. Its ability to survive in diverse environments and spread cell-to-cell makes it a major public health concern.
The p60 protein, encoded by the iap gene, is a major extracellular autolysin (murein hydrolase) essential for bacterial cell division and daughter cell separation. It is a 60 kDa protein containing LysM (for cell wall adhesion) and catalytic domains. Critically, p60 is secreted in large, stable quantities into the culture supernatant, making it an excellent, accessible target for immunological detection assays, unlike strictly intracellular proteins.
Beyond cell division, p60 is a significant virulence factor, contributing to the pathogen's adherence, host cell invasion, and sustained colonization. Its fragments can also modulate the innate immune response, such as stimulating Natural Killer (NK) cells.
