- Lymphocyte-activation gene 3 (LAG3), also known as CD223, encodes the inhibitory receptor lymphocyte activation gene 3 protein, binding to ligands, such as FGL1, and forms conserved signal interactions.
- As an immune checkpoint, LAG3 is expressed primarily in activated T-cells and a subset of natural killer (NK) cells in tissues, such as the spleen and lymph nodes.
- Interleukin-2, interleukin-7, or interleukin-12 induces the expression of LAG3 on activated T-cells. By binding to its ligands, LAG3 transmits an inhibitory signal, which further affects the immune function.

LAG3 Ligands
LAG-3 distinguishes and binds to pMHCII. Galectin-3 and LSECtin interact with LAG-3 on glycosylated sites and inhibit the IFN-γ production from T cells. FGL1 contributes to the inhibitory function of LAG-3 by reducing the secretion of IL-2.
