Introduction of L. pneumophilia

Legionella pneumophila (L. pneumophila) is a ubiquitous Gram-negative bacterium found in freshwater environments, where it survives inside free-living protozoa. When aerosolized and inhaled by humans, it causes severe pneumonia known as Legionnaires' disease, or the milder Pontiac fever, necessitating robust detection methods.

Its pathogenicity depends on its ability to infect and replicate within host alveolar macrophages. Upon being phagocytosed, L. pneumophila prevents phagosome-lysosome fusion and creates the Legionella-containing vacuole (LCV). This process is mediated by the Dot/Icm Type IV secretion system, which injects hundreds of effector proteins to hijack host cell processes (including trafficking and signaling) for bacterial growth. Diagnostics primarily rely on studying the bacterial surface structures, particularly the highly immunogenic Lipopolysaccharide (LPS) from serogroup 1.
