Myostatin (MSTN), a secreted member of the transforming growth factor-beta superfamily, acts as a ligand. Within this family, ligands bind to diverse TGF-beta receptors, initiating the recruitment and activation of SMAD family transcription factors that govern gene expression. MSTN undergoes proteolytic processing, resulting in the generation of individual subunits that form a disulfide-linked homodimer. It exerts a negative regulatory influence on the proliferation and differentiation of skeletal muscle cells, impeding the development and growth of skeletal muscle through paracrine or autocrine signaling.

MSTN conveys signals to the nucleus through a series of consecutive reactions. The MSTN dimer initially binds to ActRIIB, facilitating the downstream Smad protein pathway, consequently blocking Smad-mediated MSTN signaling. Nevertheless, MSTN signaling also diminishes Akt/TORC1/p70S6K signaling, hindering myoblast differentiation and myotube size in vitro. MSTN has emerged as a prospective therapeutic target in the realm of muscular disorders, which is a significant negative regulator of muscle mass.
