Neural Cell Adhesion Molecule 1 (NCAM1), also known as NCAM or CD56, is a critical member of the immunoglobulin superfamily. NCAM1 is a glycoprotein mainly found on the surface of neurons, glial cells, and skeletal muscle. NCAM1 is also detected in T cells and dendritic cells. NCAM1 has been reported to be a critical regulator of cell-cell adhesion, synaptic plasticity, and neurite growth.

NCAM1 is a crucial regulator of cell-cell interactions and cell-matrix interactions during development and differentiation. NCAM1 participates in the development of the nervous system by regulating neurogenesis and neurite outgrowth. Furthermore, NCAM1 plays a crucial role in a variety of brain-related biological processes, including neuronal migration, axon branching, and synaptogenesis. In nerves, NCAM1 regulates homogeneous interactions between neurons and muscles. It is worth noting that NCAM1 is also a key regulator of the expansion of T cells and dendritic cells, which are closely associated with immune surveillance.
NCAM1 is reported to signal through interactions with N-cadherin, fibroblast growth factor receptor (FGFR), and other components of the extracellular matrix, triggering signaling cascades involving MAPK, FAK, and PI3K. NCAM1 also acts as a signal transduction receptor molecule and is involved in regulating various biological phenomena, including cell adhesion, proliferation, and survival.
