PVR (poliovirus receptor), also known as CD155 or NECL5, is a vital member of the immunoglobulin superfamily. As a typical transmembrane protein, PVR contains 3 specific domains, one of which can be recognized by the virus. Therefore, PVR is also a crucial mediator for poliovirus to enter cells. The main function of PVR is to construct intercellular junctions.

Functions

As a vital adhesion molecule, PVR plays a pivotal role in many physiological processes, such as cell migration and proliferation. Data show that PVR may also participate in the humoral immune response in the intestine. In addition, PVR is found to be overexpressed on tumor cells, and contrary to the pro-tumor function of some proteins, PVR is involved in anti-tumor immune responses. PVR also serves as a stress-inducing molecule that deactivates danger signals, alerting the immune system to prevent neoplastic transformation.

PVR Pathways

PVR is reported to accelerate the cell cycle via the Ras/Raf/MEK/ERK pathway. Specifically, PVR enhances Ras/Raf/MEK/ERK signaling, leading to changes in the expression of cell cycle proteins and thereby regulating the cell cycle. Furthermore, evidence reveals that the Ras oncogene mediates the upregulation of PVR through the Raf/MEK/ERK/activator protein 1 signaling pathway.

PVR and Diseases

A large amount of evidence shows that PVR is abnormally expressed in many tumors. In addition, PVR protein levels are closely linked to tumorigenesis and poor prognosis. Data show that serum levels of soluble PVR in cancer patients are significantly higher than those in the normal population. Based on these facts, PVR has gradually emerged as a potential marker for assessing cancer progression and prognosis.
