TNFSF8, also known as CD30 ligand (CD30L), or CD153, is a 26-40 kD type-II glycoprotein membrane belonging to tumor necrosis factor family members (TNFSF). It is expressed on activated CD4+ and CD8+ T cells, monocytes/macrophages, granulocytes, and epithelial cells in the thymic medulla and in some Burkitt's lymphoma cell lines. The cross-linking of cell surface TNFSF8 by specific antibodies or soluble CD30 can transduce signals to induce granulocytes and T cell activation, this suggests that TNFSF members and their cognate receptors may be signaled bi-directionally. The human TNFSF8 gene is located at 9q33. TNFSF8 has an extracellular domain of 172 amino acids and a cytoplasmic domain of 40 amino acid residues.

TNFSF8 in Signaling

TNFSF8 activates key signaling pathways mainly through interaction with its receptor CD30, including:

  • NF-κB signaling pathway

TNFSF8-CD30 interaction activates the nuclear factor kappa B (NF-κB) signaling pathway. NF-κB serves as a pivotal transcription factor overseeing the expression of numerous genes integral to immune response, inflammatory response, and cell survival.

  • MAPK/ERK signaling pathway

TNFSF8 can activate the mitogen-activated protein kinase (MAPK)/extracellular signal-regulated kinase (ERK) pathway through CD30. This signaling pathway is important in cell proliferation, differentiation, and survival.

  • JNK signaling pathway

The Jun N-terminal kinase (JNK) pathway assumes a critical role in cellular stress response, inflammatory reactions, and apoptosis.

  • PI3K/AKT signaling pathway

The TNFSF8-CD30 interaction can activate the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (AKT) pathway. The PI3K/AKT pathway plays a key role in cell growth, survival, and metabolic regulation.

TNFSF8 Antibodies in Therapeutics

Increasing evidence supports the pivotal role of immunosenescence in various age-related chronic disorders and cancer. Senescence-associated T (SA-T) cells, characterized by impaired proliferation and the secretion of senescence-associated secretory phenotype (SASP), are among the earliest T cell populations demonstrating classical senescence features. The interaction between TNFSF8 and CD30 is crucial in driving the reciprocal activation of SA-T cells, contributing to immunosenescent phenotypes and autoimmunity. Antibodies targeting TNFSF8 mediate the blockade of TNFSF8/CD30 interaction, which suppresses the age-related increase in SA-T cells, thus improving autoimmune diseases such as lupus symptoms.
