Product listVISTA (V-domain Ig Suppressor of T cell Activation), also known as VSIR, is a novel inhibitory immune checkpoint protein. VISTA is expressed on the surface of the cells, such as myeloid-derived suppressor cells (MDSCs), tumor-infiltrating lymphocytes (TILs), and tumor cells. VISTA is pH selectively interacted with ligands such as PSGL-1 and VSIG3. As an immunoregulatory receptor, it plays a critical inhibitory role in regulating immune responses, particularly in solid tumor immunotherapy.

VISTA Signaling & Interaction & Function
VISTA is found in various tissues and in various cancers. Its expression, regulation, and function may differ in a complex way in different cancers. Here are some interactions in which VISTA may be involved:
- Inhibits MyD88-dependent TLR signaling and production of proinflammatory cytokines and mediators (Fig. 1B).
- Interacts with VSIG3 and subsequently suppresses T cell activation and expansion (Fig. 1C).
- Binds to PSGL-1 at acidic pH with uncharacterized signaling pathways (Fig. 1D–F).
- Inhibits TCR signaling and cytokine production and promotes T cell exhaustion.

Clinical significance
- For solid tumor immunotherapy, mAbs against VISTA with blocking or neutralizing functions are currently in preclinical or clinical trials, as listed in Table 1.
- For diagnostic and prognostic research, anti-VISTA mAbs could quantify the existence of the negative immune checkpoint protein VISTA, which is a risk factor for cancer metastasis and recurrence.
